Both cognitive aging and Alzheimer’s disease (AD) are characterized by age-related deficits in performance on specific cognitive domains. In line with current theoretical models, both clinical and imaging studies have suggested that the deficits in AD may arise from a dysfunctional relationship between cognitively “older” and “younger” brain regions, with the latter becoming relatively inefficient due to loss of functional synaptic connections. Despite these similarities, however, little research has examined whether the cognitive symptoms of AD may be explained by a general age-related decline in the efficiency of brain regions. To address this issue, we examined whether the cognitive deficits of AD patients are best characterized as primarily resulting from an age-related decline in functional brain connectivity or whether they may instead be attributed to a more general loss of efficiency. Our approach was to consider the role of the Default Mode Network, a well-characterized, brain-wide resting-state network that appears to decline in functional connectivity with increasing age. Specifically, we considered whether Default Mode Network connectivity may play an important role in cognitive aging, and whether AD patients may exhibit an increase in Default Mode Network connectivity, or a reduction in the decline in Default Mode Network connectivity with increasing age, compared with healthy older adults. Data were collected from 36 healthy older adults, 36 patients with AD, and 16 age-matched controls. Our findings suggest that Default Mode Network connectivity is not sensitive to age and, therefore, not likely to contribute significantly to age-related cognitive changes. Moreover, based on a recent meta-analysis, we conclude that the Default Mode Network does not decline in functional connectivity with increasing age, and we suggest alternative interpretations for the cognitive deficits of AD patients.Biden: ‘Everything in this Country is on the Table’
